If I drill through a 2x4 using a diamond hole saw (and cooling with water), will it ruin the bit?

No, but it will be a rough cut in the wood due to the tooth's design. Those blades are designed to cut other "stone" materials. Use a Carbide-tipped saw blade for wood

Thanks, just being lazy and didn't want to buy another bit.
As long as the hole works as a guide for glass cutting, I don't care how rough it is lol

Yeah you can. I’ve done it. It’s going to dull the diamond bit quicker but it still works. Your best bet is to buy a metric wood hole saw that matches.

The real issue is that the diamond bit lacks a pilot. You could just as easily cut the glass as you could the wood. The point of a wood hole saw is that you can created a precise template that the diamond bit can work within.

i was only curious. I have a full set of hole saws so “cutting“ wood with a diamond anything is a predicament that hopefully will remain something of lore.

Well, I did it - I used a piece of hardboard (think pressed fibers to be wood-like) as a guide. The diamond drill was noticably dulled. I had cut one small aquarium, then the board, then a larger tank. It worked, but barely. This is an old post - but someone still needs to drill a tank.
